Sure, any save (from "kill" effects) can be used against Tears of Lys.

should be an easy answer

 

tears of lys:

after you win an int challenge as the attacker place a poison token on a character without int controlled by the losing opponent. at the end of the phase, if that character still has a poison icon remove it and kill that character. limit once per challenge

 

I'm assuming duplicate can save the character, in which case tears isn't as good as I thought.

And the token is removed or not? Since the interrupt comes before the effect that kills the char i'm not 100% sure it does

A dupe's save doesn't cancel or undo any part of the initiation of the effect. It only stops the character from dying when the kill is resolved. Any other part of a "kill and..." effect will take place normally.
