I think the text is wrong, it should be: "At least one opponents defending character will be burned in each challenge."

If I declare a challenge and my character is killed before my opponent declares defenders, does he still has to defend with someone (since the challenge has been launched anyway)?
Yes, after a character is declared as an attacker - even if it dies - the challenge was initiated and must resolve. Timing is described in detail here.
