After Gondorian Spearman (Core) killed an attacking enemy, is he still considered a defending character to use this card?
Yes. Once he is declared as a defender, he is considered a defending character until the end of the resolution of that attack. There is a player action window after you declare defender(s), in which you can use this card on the Spearman to ready him. Even if you have already triggered the Spearman's response, he is still defending.

Hmm does this mean you can profit from the +1 defense during the enemy attack you originally used as the trigger for this card? (haven´t thought it might be used like that before)
The strong part is readying youe character which can be a hero or an ally too
