Great identity and is great for early-mid game mind games.
"Ignore this ability until the end of the turn whenever the Runner runs on a central server"

does he have to succeed?
No, simply starting a run and dropping out at the first opportunity satisfies the requirement.

However, it's noteworthy that the Runner cannot jack out of the run while approaching the first piece of ice protecting a server, so if it's something like a Neural Katana, you might be able to make them spend credits even if they have no intention of making the run succeed.

Just make sure the outermost piece of ice on each server hurts!
This manipulates runners so bad - the possibilities are endless. First use - in conjunction with a deliberately accessible Snare! or two.
It's soooo OP!
But I don't play jinteki XD

No, he can jack out after encountering the first ice.

Seems like now isn't such a bad time to start.

I've been using this identity with my first deck. It's nice in that it forces the runners to ration two clicks in order to run at a remote. On the other hand... I feel like it causes me to run a very ice heavy deck, and spread my ice thinner than I might with the other identity.
RULING:
Run deflected from central to remote with Bullfrog still counts for this IDs condition. So Bullfrog doesn't help this ID.

If I end the run with Nisei Mk II as soon as it's declared, is the condition satisfied?

No because they still made a run at one of your central servers (it doesn't have to be successful, heck I do it with snitch then jack before even seeing ICE), besides why wouldn't you wait till they have tried to run both servers and only if they are about to access an agenda Nisei it?
