Is the "When revealed" of a plot card...say A City Besieged...a triggered effect?
No, it is a passive effect. Triggered effects always say the phase, or Any Phase, in bold, before giving the effect.

If you have a lot of unique characters, this card is great against Burn.

No as NuFenix said, but it's also worth pointing out that A City Besieged has two targets not one, so it wouldn't work in that case anyway ;).

Yes. Anything that is precceeded by bold text, such as Any Phase, Challenges, Response, etc is considered a triggered effect.

Does this card cancel Varamys Sixskins effect?

No. Varamyr doesn't choose/target anything.
