Can this be used to reduce the additional cost from City of Shadows (CoS) when using out-of-house shadows cards?
Yes it can. Just like if you are playing an out of house character, you can use reducers to reduce the out of house penalty there too.
Hidden Chambers isnt unique. Will two of them reduce the costs by 2?

Yes and no. If you are bringing them out of shadows during the normal shadows phase event, then yes it will reduce them, but if you are using their ability, you must pay the full cost to do so, and thus Hidden Chambers can't be used.

Are you sure? I've heard that since this reduces the cost it would work with Meera and Wolf. . .

I think the distinction is more in the fact that Hidden Chambers is used to reduce the cost of bringing something out of shadows, when done during the standard Shadows action during the beginning of the phase framework.

Meera and the Guardian Wolf are both triggered effects that have a cost. That cost is specified in the ability by forcing a payment of the printed cost to bring them out of shadows.

I do see the arguement presented that you are still "bringing a card out of shadows" so why wouldn't the Hidden Chambers effect apply...

However, 1 is "bringing a card out of shadows" and the other is "triggering a character ability that happens to bring that card out of shadows"

Though I don't have a specific ruling in mind for this conclusion - I suggest hitting up the FFG forums.
