What does it mean by "all instances of all challenge icons"?
"Loses all instances" effectively means the character can never gain them back, no matter how many "+1 icon" effects are applied.
Does this effect persist even if the character leaves play and somehow comes back into play?
No. When a card leaves play and returns, it is considered a new card - even if it is the same piece of cardboard. That, taken along with the fact that cards are only affected while they are in play (unless something specifically says otherwise), means that all lasting effects on a card end when it leaves play. (How awful would Narrow Escape be against burn effects if things didn't work this way?)
Ok, fair enough. I guess the wording "until the end of the game" threw me a little. Is there a reason why it was worded this way, rather than just "That character gains Prized 1 and loses all instances of all challenge icons"?
Because if you just said, "That character gains Prized 1 and loses all instances of all challenge icons," people would argue about whether the duration was left off in error, or whether it really was permanent.
