Could you put this on a enhancement that says this is considered an objective? don't think dark side has a enhancement that says that thou.
The Light Side also doesn't have an enhancement that says it is considered to be an objective. So, to answer the theoretical question about an enhancement that does not exist... it depends. They definitely could design an enhancement worded in such a way that Defense Upgrade could be played on it.
Yes, Missions are objectives for the purposes of cards that refer to objectives. Defense upgrade in particular can be played on any objective, including your opponents.
