THE challenge means the one after kneeling meereen?

Nope, because it can only target a "participating" dragon, it has to be done during a challenge. So the challenge refers to the current challenge in which the Dragon is participating.

can I kneel this after my opponent has targeted one of my dragons with....i dont know....Die by the Sword (LoW) or something along those events?

No, as it is not a response with a save/cancel effect. You need to use this on them in advance, during that challenge.
