So, if they both participate, that's 2 power to Jon? Or does this resolve once, using either one as a trigger?
You would only get one power, as you can only trigger the Response once per thing that triggers it, one of those two characters winning a challenge.
So if you win 2 different challenges, one with Jon and one with Ghost, you get the 2 power?
Yes :)

And if you win a challenge as a defender Jon Snow still gets one power token?

Correct.  Note that with the way it is worded, if you win a challenge in which your opponent's Ghost participated, you can claim a power on Jon.  Also note that since Jon is self-referential, it does not work with your opponent's Jon Snow.
