Is a choosen HoD location (lets say Gates of Winterfell) immune to River Blockade?
Does the Iron Throne played by this agenda get affected by other Iron thrones and cancel its effect?
Kennit - Only the location gets the immunity, any abilities on the card are still vulnerable and can be cancelled.
Nikolas - The Iron throne only gains immunity to opponents cards from HoD; its not immune to its own text, so opposing thrones still shut each other down.
The apprentice is not a plot effect so yes the hod location is immune to it.
I'm surprised I haven't seen any complaints about this card. There are no drawbacks, but there are loads of benefits:
1. Search your deck for a key location at the start of the game
2. Put it into play for 2 gold, regardless of how expensive it is (so most players will be saving at least 1 gold)
3. The chosen location is then immune to opponents' non-plot card effects.
Drawing one less card for setup and only spending up to 3 gold are not drawbacks because you've already drawn the extra card and spent the extra gold.
I really think this Agenda would've been more balanced if there was more of a drawback, such as "You cannot play cards during setup" (a la KotHH), or a more severe draw restriction. I completely understand why people like to use the card, but I feel like there should be some kind of trade-off when selecting an Agenda card. This one is just too easy.
I think the trade-off comes with building. Drawing one less card and only being able to spend 3 gold means you should bring your gold curve down and your character count up if you don't want to be behind on characters turn 1-2. That and giving up the opportunity to run another agenda/no-agenda tech limits yours options.

When I use HoF to protect a location of mine, is it only protected from being discarded from play, or is its response also protected from cancel responses? I suppose what counts for HoD protected locations coutns also for Outfit for War attachment.
Short answer: immunity means that the card cannot be targeted by and ignores the direct effects of whatever it is immune to. That covers a lot more than simply "cannot be discarded." However, only the card itself is protected - power, attachments, abilities, etc. can still be affected by whatever the card is immune to (so HoD does not turn your chosen location's effects into "cannot be canceled").

Do people who play this with a card central to the deck (say, Lanni Tunnels) generally have additional protection for the location? Dupes, location recursion, etc? Or is it better to just accept that certain decks (Cities) will probably nuke your primo location eventually, and play around it?
Huh? How would it not be immune to Frozen Solid? Frozen Solid is a "non-plot card effect" isn't it?
The second copy played from hand wouldn't share immunities, but if it was played while the original was already out, it attaches as a duplicate, and can save the original, immune card from being discarded/returned to hand, yes?
If you chose The Scourge, but had all kinds of other icon removal effects in the deck, then seeing it discarded wouldn't be too much to bear. Annoying, but not disastrous. Building a deck that can still do its thing without The Scourge isn't too difficult. I'm not so sure how easy that would be with the Tunnels, though...
About the only other things hitting Hod location are Rally Cry (a risky plot to run) and Shield Islands - though chances are you steal it back it does remove the protection.
Search and Detain?
